Between Frankfurt and Venice? The entire f-ing Alps!
Anyway, I did an Alps roadtrip during last summer. I particularly liked the San Bernardino pass. It was great to drive and, unlike other passes in the Alps, the roads were close to deserted. Note that it's the twisty yellow road marked with 13, not the orange more highway-like E43.

If you are going through Munich, Hit up Fussen and the Romantic Road. You'll end up in Fussen, at the base of the Alps, with Castle Neuschwanstein near by.

As duurtlang said, crossing the Alps can be the best part. Just find a route that keeps you off the motorways and you will have fun/feel like Hannibal.
I live in south-eastern Bavaria so I could give you tips on where to go, but your request is kinda vague.
If you are starting in Frankfurt (I assume it's Franfurt/Main. There's Frankfurt/Oder close to the Polish border as well.) try to avoid the A8 between Karlsruhe and Munich. Not many unrestricted parts there. The route along Würzburg, Nürnberg, Munich on the A? and the A9 is longer but can be faster, and mos def will be more fun.

You could always take the classic route through the Netherlands, Belgium and France.

Many great roads ahead! First of all make sure you're traveling through Switzerland to Italy. There you find:
- Nufenenpass (my favourite)
- San Bernardinopass
- Gotthardpass
- Furkapass
In Italy try to take some routes near the great lakes (Maggiore, Lugano) like the SS34 from cannobio to Verbania. There you can take the ferry to the other side of the Lago Maggiore. Or, if you have the time, first take the SS631 from Cannobio to Masera an head to Lago Orta through the SS34/SS229 (Orta San Giulio).
And, if you have even more time, try to get to Monterosso al Mare from Genova. I believe it's the SS1, especially from Sestri Levante it's getting awesome.
Croatia has great highways. Lot's of good tarmac, few traffic.
On the way back to Frankfurt make sure you take the Stelvio.

Assuming you will come through Munich Area (or around it) and go south of it into Austria: If you have enough time:
1. Ötztal -> Timmelsjoch
2 Jaufenpass to Vipiteno /Sterzing
3 on to Pennserjoch until Bolzano/Bozen
4. Round the Sella (Sass Pordoi, Passo Sella, Passo Gardena, Passo Campolongo)
5 Now you're fed up with pass roads for some days :)
